From what I've read they'll get better on their own, it'll just take longer.
Can I ask why you don't want to give her medicine? Are you trying to get her to leave?

just so you know, from guides and such I've seen it seems that your friendship level with a villager has just about no impact on whether they move. i'm pretty sure the villager who gets the thought bubble is completely random (at least that's what I've been told from a few sources)

Agreed. I haven't had anybody ask to move in NH so I don't know how it works yet, but for NL, I actually had villagers who had higher friendship with me seem more likely to ask to move than villagers with lower friendship. So I never understood it when people said they hit villagers with nets or ignored them to try to make them move.
It might not even high or low friendship at all. It might just be random.

apparently after the welcome amiibo update they stopped getting sick
Either you were really lucky or you didn't care enough about your villagers to check on them inside their homes during their time of need. They definitely could get sick in NL.

I believe with the Welcome Amiibo update they got rid of the system. In vanilla new leaf they could get sick.

They never got sick, as stated, because I purchased the physical game that had Welcome Amiibo installed.
