So I see reserves are kind of a maybe thing. If someone is reserved but the person does not pick up, can we ask to be next in line or shall we just wait until the villager is announced available?

It depends how they are reserved.
If I've reserved it on a friend level, or because the villager is not really that popular, then I will lift it after a certain amount of time.
Autoreserved villagers (Where you trade one villager of yours for mine) get a day to be picked up, and will only be given away if someone else offers to trade the villager in for another of theirs, or the villager will be raffled.
