Villager wants me to catch a very hard to catch fish

He wants a blowfish and it seems rare and catchable very late. I promised to give him one.

Will he be mad if I fail to keep my promise?

Also next time he does ask and I say no, is that better, I always get the feeling they might be mad as well then.

If you don't give it to him, he won't do anything, he won't even ask about it.

The request goes away the next day. I think that saying yes and not following through doesn't damage your friendship, it's just that catching that fish increases it. My villagers ask me to catch bugs and fish for them a lot, and I always say yes but never do it unless it fits their house and I'm not planning on adopting them out. I've never noticed a big difference in my friendship levels with them.

I'm not sure about this, but I think that when you make plans to hang out with them and don't show up, that does decrease your friendship level. When you see them again, they usually say something about how they've waited for you but you never showed up.

You can always take notes when they make plans with you (you can use the note-taking app on your 3DS without quitting the game) so you won't forget. If you're planning on quitting the game before that hour, you can set the clock forward and hang out with them before you do that. When I do the latter, I usually only change the clock back the next day, because I've noticed that going back within the same day after visiting or being visited by a villager will sometimes make it seem like it didn't happen at all. Whenever I played the same day over and over to keep villagers in boxes, I noticed that sometimes the same villager would ask me to visit me twice as though they hadn't done that earlier and I wouldn't receive as many letters thanking me for hanging out with them.

Not really. :( The only way to tell is picking up their cues. As their friendship levels increase, their dialogue starts changing slightly to reflect that. The personalities that are rude to you at first become much nicer. Villagers you have a good level of friendship with will sometimes send you letters or give you gifts for no reason. When you're at a high level, they may surprise you by visiting you when you're home or give you their picture. If you own the megaphone, if you call them when you can't see them, villagers you're close to will greet you differently. At times they might also walk away from a conversation with you looking really happy and singing.

It depends on the villager and their personality and it's sort of random. For example, a villager I only started being good friends with a few days ago has already sent me her picture and visited me, while with others that I've talked to more often, it's taken longer for them to do that and sometimes they haven't done that at all.

Don't worry, everyone was new to AC or ACNL once. :)
 
Those days back I was carrying a rococo clock and Lopez stopped me to buy my item, I refused because I didn't want to screw his house, and then he got depressed and asked me to leave (╯︵╰,)

I've never had villagers refuse to talk to me after I denied them items, I had no idea that could happen. Are you sure you hadn't been talking to him a bunch of times in a row? They do that if you talk to them too much.

It is. They said they refused a trade with a villager and that villager got depressed and asked them to leave. That's what villagers say when they refuse to talk to you.

I'm sorry that you're desperate for TBT.
