It shouldn’t. Your birthday is separate event so once it’s completed, it resets to a normal day.

Isabelle should announce both events and even if she doesn’t, it should still be there.

No. You're birthday works like K.K.'s first concert. It's just an interactive cutscene that plays when your start the game.
