My main-island copy of Animal Crossing: New Horizons is digital.

I want to change the microSD card. (I used up Photos with the current card I want to replace.)

As I have inserted the new card, and am about to start ACNH, I receive the following message:

“This software is not currently on the console. Please redownload the software to continue. If the software was originally downloaded to a microSD card, please cancel, turn off the console, and insert that microSD card.”

I purchased this first copy of Animal Crossing: New Horizons as digital. It downloaded to this console on March 20, 2020 at 12:00 a.m. ET.

So, this message has been concerned—well, fearful—that if I choose “Download,” I would lose my first island. (I’m not willing.)

What I have done so far is remove the replacement microSD card and returned to the first card. (Meanwhile, its photos are up to capacity.)

Is there anyone who can help?

(Thanks!)

Redownloading the game shouldn't affect your save file in any way. The Switch gives you a warning if you're going to delete any save file, so it wouldn't just do something that'd make it disappear without youe knowledge.

I don't really know a much about the topic, but maybe the Switch moved the game file from console to card to free some space? There should be a setting to move files from card to console too

Sorry if this isn't helpful but you said you wanna move it because your card is full of storage with photos
Can't you move over photos to a another device and then delete the photos on switch to get back space?
If you own a USB cord you can connect to your switch and plug the other part in to a computer, make sure you aren't using any software at the time, go to settings on switch then go to data management, go to manage screenshots and videos and choose copy to a computer via USB connection, as long as the cord is plugged into both devices then you can get all your switch photos crystal clear on your computer and you can find them in your files, be warned if it's unplugged the photos will disappear from your computer files until you repeat this again, it doesn't permanently save the photos to your computer this way.
Once you get it on to your computer you could upload them to some kind of photo saving device or you could find somewhere to post them then right click and choose download image on them to turn them into permanent files on your computer to be safe so you can now delete them from your switch to get back free storage.
There is also an option to move data to the microSD back to the switch system storage or verse vista, unfortunately I've never done this or have any knowledge on that so I don't know how it will turn out, maybe someone else knows.
You could also archive software to free up space if your switch is overflowing the storage, I'm not sure how to do that either but I'm sure someone else knows how to archive software or there is a video on Youtube on it.
I'm aware this most likely isn't helpful I just wanted to offer advice if you said you need to replace the card only because it's storage has been filled up with photos I wanted to suggest ways to get more storage without deleting data you possibly want to keep.
I'm not sure of the answer to your original question.
