When events say that you can't edit your post after submitting, does this mean that any post that has been edited will be disqualified? Or does it just mean that any changes after the original post was made won't be taken into account when it's time for judging or deciding favorites? Or what?
I'd say this depends on the event and/or host(s), and the severity of the situation. The general concept behind the rule is for you to only submit when your piece/guess is final. For example, if you submitted a piece of artwork to a local competition, you wouldn't be able to go back the next day and ask for your piece back so you could make changes.
We would advise you to simply not edit your submission for any reason without the host's consent first.

Can we print the colouring page in the Forest of the Lustrous and colour it in with pencils/pens and scan the page to post?

If there is a rule within the event or contest thread that mentions not to edit your post once your entry is submitted, then yes, you should have the host's consent prior.
I noticed a discrepancy for the start date of Leif's Labyrinth. The thread says it starts on the 27th, but the tracker/calendar says it starts on the 20th. Which start date is actually correct?
Oops -- it should be the 20th! Fixed now. The 27th was an old date that I forgot to change.
Not sure if this has been asked yet, but will the Celeste plushies restock after the first round? I was hoping there'd be another restock for those in different timezones who can't catch the 5PM EDT opening. :]
